Construction of Polymer–Protein Bioconjugates with Varying Chain Topologies: Polymer Molecular Weight and Steric Hindrance Effects

摘要: We report on the fabrication of well-defined polymer-protein bio- conjugates with varying chain architec- tures, including star polymers, block copolymers, and heteroarm copolymers through specific nonco- valent interaction between avidin biotinylated synthetic polymer precur- sors. Homopolymer diblock pre- cursors site-specifically labeled a single biotin moiety at termi- nal, middle, or junction point were synthesized by combina- tion atom-transfer radical polymeri- zation (ATRP) click reactions. By taking advantage molecular recogni- moiet- ies, supramolecular successfully fabricat- ed. This binding process was also assessed using diffraction optic technology (DOT) technique. further investigated effects poly- mer weights, location functionality within chain, conforma- tions, that is, steric hindrance effects, numbers chains per bioconjugates, which determined standard avidin/2-(4-hydroxyazobenzene)benzoic acid (HABA) assay. The num- bers vary in range 1.9-3.3, de- pending lo- cations precursors, conformations.
