摘要: The gene encoding the 54 kDa protein of signal recognition particle (SRP54) in hyperthermophilic archaeon Pyrococcus furiosus has been cloned and sequenced. Recombinant P. SRP54 (pf-SRP54) N-terminal G-domain C-terminal M-domain (pf-SRP54M) pf-SRP54 with an amino-terminal addition six histidine residues were expressed Escherichia coli subjected to binding experiments for SRP RNA, non-conserved 213-nucleotide RNA (helices 1, 2, 3, 4 5) conserved 107-nucleotide 6 8) from RNA. properties purified determined by filter assays. histidine-tagged pf-SRP54M bound specifically absence pf-SRP19, unlike eukaryotic homologue, apparent constant (K) 18 nM.
