Prognosis-related molecular markers in pediatric central nervous system tumors.

摘要: In the wake of recent progress in understanding genetic pathways involved development brain tumors, a major goal is to correlate molecular data with clinical outcome, survival, and response treatment modalities. This particular importance among pediatric population. Reliable prognostic factors could potentially permit tailoring therapy that only patients most aggressive tumors would receive intense treatments. A survey publications about prognosis-related features revealed 74 series, which 46 presented statistically significant outcome-associated parameters as defined by p value <0.05. Most investigations revealing were performed on medulloblastomas (34 publications), followed astrocytic (6 publications) ependymomas (5 publications). Promising approaches markers include gene expression profiles, DNA ploidy, loss heterozygosity chromosomal aberrations detected CGH FISH (1q, 17p, 17q), well oncogenes/ tumor suppressor genes their proteins (TP53, PTEN, c-erbB2, N-myc, c-myc), growth factor hormonal receptors (PDGFRA, VEGF, EGFR, HER2, HER4, ErbB-2, hTERT, TrkC), cell cycle (p27) adhesion molecules, related therapeutic resistance (multi-drug resistance, topoisomerase IIalpha, metallothionein, P-glycoprotein, tenascin). review discusses predictive potential for outcome influence decision-making children tumors.
