NMDAR Channel Segments Forming the Extracellular Vestibule Inferred from the Accessibility of Substituted Cysteines

摘要: In NMDA receptor channels, the M2 loop forms narrow constriction and cytoplasmic vestibule. The identity of an extracellular vestibule leading toward remained unresolved. Using substituted cysteine accessibility method (SCAM), we identified channel-lining residues NR1 subunit in region preceding M1 (preM1), C-terminal part M3 (M3C), N-terminal M4 (M4N). These are located on side and, with one exception, exposed to pore independently channel activation, suggesting that gate is at or further it. Permeation Ca2+ ions was decreased by mutations M3C M4N, but not preM1, a functionally distinct contribution segments channel.
