Evaluation of inter-batch differences in stem-cell derived neurons.

摘要: Differentiated cells retain the genetic information of donor but extent to which phenotypic differences between donors or batches differentiated are explained by variation introduced during differentiation process is not fully understood. In this study, we evaluated four separate commercially available neurons originating from same iPSCs investigate whether used in manufacturing affected genome-wide gene expression and modified cytosines, neuronal sensitivity drugs. No significant changes expression, as measured RNA-Seq, cytosine modification levels, Illumina 450K arrays, were observed relative over time. As expected, neurotoxic chemotherapeutics outgrowth, no inter-batch paclitaxel, vincristine cisplatin. a testament utility model for studies neuropathy, that genes involved neuropathy had relatively higher levels these samples across different time points. Our results suggest differentiate into consistent, resulting minimal intra-individual variability batches. Therefore, reasonable human druggable targets prevent other neurological diseases.
