摘要: Carbonic anhydrase catalyzes the reversible hydration of carbon dioxide to bicarbonate and maintains balance CO2/HCO3- in intracellular environment, specifically for carboxylation/decarboxylation reactions. In Corynebacterium glutamicum, two putative genes, namely bca (cg2954) gca (cg0155) coding β-type γ-type carbonic anhydrase, respectively, have been identified. We here analyze transcriptional organization these genes. The start site (TSS) gene was shown be first nucleotide “A” its translational codon (ATG) thus, codes a leaderless transcript. TSS identified as an residue located at position -20 relative annotated cg0154 gene, which is immediately upstream gca. Comparative expression analysis revealed source-dependent regulation with 1.5- 2-fold lower promoter activity cells grown on acetate compared glucose sole source. Based higher mutant deficient regulator metabolism RamA wild-type C. glutamicum based binding His-tagged protein region, we present evidence that negatively regulates glutamicum. Functional characterization deletion same growth characteristics ∆gca no effect gene.
