摘要: The human 5-hydroxytryptamine 5-HT1A receptor gene was transfected into Chinese hamster ovary cells. A series of recombinant monoclonal cell lines expressing the were isolated and properties one line that expressed receptors at a high level (2.8 pmol/mg) studied in detail. In ligand binding assays with selective agonist 2-(NN-di[3H]propylamino)-8-hydroxy-1,2,3,4-tetrahydronaphthalene ([3H]8-OH-DPAT) only single class saturable high-affinity sites detected, pharmacological profile competition experiments essentially identical to bovine hippocampus. [3H]8-OH-DPAT membranes inhibited by GTP, showing cells couple G-proteins. agonists forskolin-stimulated adenylate cyclase activity and, despite expression, their apparent efficacies similar those observed for inhibition brain. This provides complete model system studying its transmembrane signalling system. can also be grown suspension culture long periods but, whereas numbers regulation guanine nucleotides are maintained suspension-grown cells, is gradually lost.
