摘要: The combination of ultraviolet absorption and circular dichroism (CD) spectroscopies provides a sensitive technique for following protein conformation conformational changes in solution. It is reasonable to predict that the application these methods study biomembrane structure should yield valuable information about their averaged might occur with functional state. And indeed they do, after only appropriate data analyses, particulate nature membranes gives rise correctable scattering artifacts. In considering CD spectrum membranes, objective eliminate distortions due by correcting absorbance or difference values would have occurred had membrane proteins been molecularly dispersed state conformationally equivalent membrane. corrected then compared reference spectra estimate average and/or different states follow possible changes. If suspension challenged, example, addition substrate one membrane-bound enzymes, its observed, this cannot be judged real until problem has properly analyzed. This not an onerous task, straightforward, large measure arguments known as pseudoreference (prs) approach described detail 1972(1) but whose origins go back 1968. (2) chapter will consider first spectroscopy related phenomenon dispersion counterpart, optical rotatory dispersion, both solutions suspensions. purple provide sample calculation demonstrating prs approach.
