Changes in the plasma proteome of Manduca sexta larvae in relation to the transcriptome variations after an immune challenge: evidence for high molecular weight immune complex formation

摘要: Manduca sextais a lepidopteran model widely used to study insect physiological processes, including innate immunity. In this study, we explored the proteomes of cell-free hemolymph from larvae injected with sterile buffer (C for control) or mixture bacteria (I induced). Of 654 proteins identified, 70 showed 1.67 >200-fold abundance increases after immune challenge; 51 decreased 0-60% control levels. While there was no strong parallel between plasma protein levels and their transcript in hemocytes fat body, mRNA level changes (i.e.I/C ratios normalized read numbers) tissues concurred spectral counts) correlation coefficients 0.44 0.57, respectively. Better correlations support that body contributes more significant portion involved various aspects Consistently, were better correlated immunity-related than unrelated ones. There is set whose apparent molecular masses differ considerably calculatedMr's, suggestive posttranslational modifications. addition, some lowMrproteins detected range 80 >300 kDa on reducing SDS-polyacrylamide gel, indicating existence highMrcovalent complexes.
