Effects of dopamine on the glutathione metabolism of cultured astroglial cells: implications for Parkinson's disease.

摘要: To investigate the effects of dopamine (DA) on release glutathione (GSH) from astrocytes, we used astroglia-rich primary cultures brains newborn rats. In absence DA, GSH accumulated in medium these with a constant rate. contrast, during incubation cells 50 µm DA extracellular was not detectable anymore. This disappearance prevented by superoxide dismutase, indicating that does affect but rather reacts released superoxide-dependent reaction. Incubation astroglial 0.5 and 1 mm established almost concentrations H2O2 5 µm 15 µm, respectively. Under conditions disulphide (GSSG). GSSG export blocked catalase MK571, an inhibitor multidrug resistance protein 1. The accumulations were modulated inhibitors receptors, transport, monoamine oxidases. other catecholamines adrenalineandnoradrenaline showed similar accumulation compared those obtained for DA. conclusion, data presented demonstrate affects metabolism two mechanisms: (i) directly chemical reaction GSH, (ii) indirectly generation hydrogen peroxide leads to efflux cells.
