Mammalian beta 1- and beta 2-adrenergic receptors. Immunological and structural comparisons.

摘要: Beta 1- and beta 2-adrenergic receptors, pharmacologically distinct proteins, have been reported to be structurally dissimilar. In the present study three techniques were employed compare nature of mammalian receptors. Antibodies against each receptor subtypes raised separately. Polyclonal antisera 1-receptors rat fat cells in mice, 2-receptors guinea pig lung rabbits. Receptors purified from (beta 1-), S49 mouse lymphoma 2-), liver 2-) probed with these antisera. Each anti-receptor demonstrated ability immunoprecipitate receptors both 2- subtypes. The mobility beta-receptors subjected polyacrylamide gel electrophoresis was using antireceptor antibodies nitrocellulose blots gels. Fat cell 1-adrenergic display Mr = 67,000 under reducing conditions 54,000 nonreducing conditions, as previously (Moxham, C. P., Malbon, (1985) Biochemistry 24, 6072-6077).
