摘要: Aberrations in membrane trafficking pathways have profound effects cellular dynamics of sorting processes and can drive severe physiological outcomes. Sorting nexin 27 (SNX27) is a metazoan-specific protein from the PX-FERM domain family required for endosomal recycling many important transmembrane receptors. Multiple studies shown SNX27-mediated requires association with retromer, one best-known regulators trafficking. SNX27/retromer downregulation strongly linked to Down’s Syndrome (DS) via glutamate receptor dysfunction Alzheimer’s Disease (AD) through increased intracellular production amyloid peptides precursor (APP) breakdown. SNX27 further addiction its role potassium channel trafficking, over-expression tumorigenesis, cancer progression, metastasis. Thus, correct multiple receptors by vital normal function prevent human diseases. The regulating cargo endosomes cell surface firmly established, but how assembles retromer generate tubulovesicular carriers remains elusive. Whether may be putative therapeutic target neurodegenerative disease now an emerging area study. This review will provide update on our molecular understanding events mediated complex endosomes.
