Early Detection of Mycobacteria Using a Novel Hydrogel Culture Method

摘要: Background: Early laboratory detection of Mycobacterium tuberculosis is crucial for controlling tuberculosis. We developed a hydrogel mycobacterial culture method that retains the advantages both solid and liquid methods in terms speed, cost, efficiency. Methods: bovis bacillus Calmette-Guerin (BCG) suspensions 200 acid-fast bacilli (AFB)-positive clinical specimens were inoculated Middlebrook 7H9 media (Becton-Dickinson Company, USA) mixed with 75 μL 9-fluorenylmethoxycarbonyl (Fmoc)-Phe-Phe-OH stock solution an Eppendorf tube just before incubation. The mixtures cultured at 37°C as long 14 days to monitor status. Results: number M. BCG increased time. For AFB smear-positive specimens, 155 158 conventional culture-positive 4 culture-negative or contaminated yielded positive cultures within days. 128 method, time using (mean, 12.6 days; range, 7 days) was significantly shorter than 16.2 6 31 P <0.0001). Conclusions: scaffold system useful timely, economical, efficient mycobacteria specimens.
