Formation of monoenoic fatty acids by desaturation in rat brain homogenate. Some properties of the enzyme system of 10-day-old brain.

摘要: Abstract Desaturation of [1-14C]stearic acid, [1-14C]palmitic and [1-14C]stearoyl-CoA was measured in homogenates 10-day-old rat brain. maximal with stearoyl-CoA at pH 6.0 to 6.6. With free fatty acids, monoene formation 7.2 7.6; stearic acid more actively desaturated than palmitic acid. Activity or linear protein concentration 2 6 mg per ml time 10 min optimal 37 45°. At 7.4, desaturation not limited by the acyl-CoA derivatives when coenzyme A, Mg2+, ATP were added. Rapid loss activity vitro prevented accurate subcellular location enzyme system. Oxygen NAD+ NADH necessary for activity. Diphosphonucleotides active triphosphonucleotides, oxidized as effective reduced forms. The fraction from labeled 91% oleic (18:1(n-9)); produced 22% palmitoleic (16:1(n-7)) 72% cis-vaccenic (18:1(n-7)). concentrations approximating normal tissue evels, these monoenes did inhibit desaturation.
