Glutamine analogs promote cytoophidium assembly in human and Drosophila cells.

摘要: CTP synthase is compartmentalized within a subcellular structure, termed the cytoophidium, in range of organisms including bacteria, yeast, fruit fly and rat. Here we show that also into cytoophidia human cells. Surprisingly, occurrence cells increases upon treatment with glutamine analog 6-diazo-5-oxo-l-norleucine (DON), an inhibitor glutamine-dependent enzymes synthase. Experiments flies confirmed DON globally promotes cytoophidium assembly. Clonal analysis via RNA interference somatic indicates expression level critical for formation cytoophidia. Moreover, facilitates assembly even when low. A second azaserine cytoophidum formation. Our data demonstrate analogs serve as useful tools study
