Locomotor Recovery in Spinal Cord-Injured Rats Treated with an Antibody Neutralizing the Myelin-Associated Neurite Growth Inhibitor Nogo-A

摘要: The limited plastic and regenerative capabilities of axons in the adult mammalian CNS can be enhanced by application a monoclonal antibody (mAb), IN-1, raised against myelin-associated neurite growth inhibitor Nogo-A. aim present study was to investigate effects this treatment on functional recovery rats with dorsal over-hemisection spinal cord. Directly after injury, half animals were implanted mAb IN-1-secreting hybridoma cells, whereas others received cells secreting control (anti-HRP). A broad spectrum locomotor tests (open field locomotor) score, grid walk, misstep withdrawal response, narrow-beam crossing) used characterize during 5 weeks injury. In all behavioral tests, IN-1-treated group significantly augmented compared antibody-treated rats. EMG recordings flexor extensor muscles treadmill walking confirmed improvement pattern rats; step-cycle duration, rhythmicity, coupling hindlimbs improved. No differences between two groups regard nociception observed tail flick test operation. These results indicating improved suggest that increased Nogo-A neutralization result functionally meaningful rewiring motor systems.
