摘要: An oxidized derivative of linoleic acid, 13-hydroxyoctadecadienoic acid (13-HODE), is dehydrogenated by an NAD+ dependent dehydrogenase present in rat colon mucosa. The product the reaction 2,4-dienone, 13-oxooctadecadienoic acid. Enzyme activity was determined HPLC analysis incubation mixtures as well measuring increase absorbance at 285 nm, which represents formation 2,4-dienone chromophore. Characteristics with respect to protein concentration, time and substrate dependence were investigated. Several inhibitors known dehydrogenases had no effect on 13-HODE dehydrogenase. These include, ethanol, indomethacin, 6-methyl-17-hydroxyprogesterone acetate, 4-(diethylamino)-benzaldehyde, aspirin. enzyme mildly inhibited pyrazole, 4-methylpyrazole ibuprofen. Disulfiram found be a potent inhibitor IC50 200 microM. Inhibitor specificity, other characteristics suggest neither alcohol dehydrogenase, diol nor prostaglandin It possible this plays important role response colonic mucosa mitogenic fatty acids.
