Maturation of the Human Complement System. I. ONSET TIME AND SITES OF FETAL C1q, C4, C3, AND C5 SYNTHESIS

摘要: The onset times and sites of human C1q, C4, C3, C5 synthesis were determined by culturing tissues from 23 fetuses, 8-25 wk old, in the presence [(14)C]lysine isoleucine. In parallel, IgG IgM production was followed. Liver, spleen, placenta, peritoneal bone marrow cells, thymus, colon cultured for 48 h concentrated media studied immunoelectrophoresis subsequent autoradiography using adult serum as carrier specific antisera. quantitative approximated scoring intensity labeled precipitin lines uniform conditions. detected earliest at 8 gestation 11 thereafter, uniformly present multiple tissues. C1q synthesis, however, limited almost exclusively to began 14 wk, not present. contrast did occur three fetuses synthesizing complement while early wk. inconstant, occurred predominantly quantitatively much less compared C5. These findings suggest that developmentally system is a more primative biological defense mechanism than antibody.
