Phytoplasma transmission by in vitro graft inoculation as a basis for a preliminary screening method for resistance in fruit trees

摘要: In vitro grafting was tested as a technique for inoculating Prunus rootstock marianna GF 8-1 with European stone fruit yellows (ESFY) phytoplasmas and apple Malus pumila MM106 proliferation (AP) phytoplasmas. shoot cultures of ESFY-infected AP-infected were used graft inoculum to transmit the respective healthy rootstock. Phytoplasma transmission assessed after contact 1, 2 or 3 months. Healthy autografts controls monitor parameters in grafting. Successful union formation ranged from 58 79% irrespective plant species sanitary state graft. Pathogen-specific polymerase chain reaction (PCR) test inoculated rootstocks presence ESFY AP phytoplasmas, respectively. The rate phytoplasma successful Prunus-grafts increased 69 94% time contact. occurred 80 97% grafts. To our knowledge this is first report by vitro. results provide good basis establishment preliminary screening method resistance Malus.
