摘要: β-Hydroxyisobutyrate dehydrogenase was purified and crystallized from Candida rugosa IFO 0750. The overall purification about 3700-fold with a yield of 34.5%. molecular weight estimated to be 80,000 by the gel filtration method 75,000 sedimentation velocity method. enzyme consisted two identical subunits weights 40,000. dehydrogenated both l- d-β-hydroxyisobutyric acid (β-HIBA) NAD+ as coenzyme. specific activities for d-β-HIBA were 36.1 8.94 units per mg protein, respectively. Km values l-, 3.70 × 10−4M, 1.25 10−3M 5.00 10−5M, It suggested kinetic analysis that dehydrogenations catalyzed at single active center enzyme.
