摘要: Cytokine secretion by endometrial cells from estrous and mated mice was measured using specific bioassays. The granulocytemacrophage colony-stimulating factor (GM-CSF) interleukin-6 (IL-6) contents of uterine intraluminal fluid were elevated> 20-fold 250-fold respectively following mating, both cytokines synthesized in abundance vitro harvested at estrus on Day 1 pregnancy. Synthesis not impaired genetically lymphocyte-deficient nude, SCID, or beige mice. To determine the cellular origin cytokines, a panning technique employing monoclonal antibodies against range leukocyte other lineage markers used to isolate cell subsets vitro. These experiments identified glandular and/or luminal epithelial as major source GM-CSF IL-6 pregnant uteri. Stromal fibroblasts also IL-6, did macrophages Epithelial midgestation uteri secreted lL-6 quantities similar those Bioactivities derived neutralized antibodies, size-exclusion chromatography conditioned media revealed peaks bioactivity with M, 23000 23 000-26 000, respectively. Bioassay fluids culture supernatantswere negative for interleukin-l, interleukin-2, interleukin-3, tumor necrosis factor-u. studies identify murine epithelium potent lL-6, which we postulate have potentially important functions pregnancy through actions target uterus conceptus.
