摘要: Abstract Detergents are important reagents in membrane biochemistry. Since each system studied places different demands on the detergent terms of desirous physicochemical properties, detergents new to biochemistry must continuously be sought. Ammonium perfluorooctanoate (APFO) was investigated, as representative fluorinated surfactants, its suitability a "biological detergent." It did not interfere with Markwell modification Lowry procedure at concentrations up 2% (w/v). Critical micellization concentration (cmc) values (0.013-0.0275 M) for this were determined number buffers biological interest. demonstrated that can removed by dialysis, albeit slowly. This slow removal may particularly useful reconstitution/crystallization studies. Solubilization studies several systems containing proteins listed (the major protein sector vacuolar H + -ATPase (16 kDa protein); photosystem II; equine herpes virus (EHV) envelope proteins) indicate it is potent solubilizing agent, likely enhance yield cases where solubilization has already been demonstrated, and, other cases, solubilize formerly recalcitrant solubilization. The APFO from solubilized 16-kDa means Extracti-Gel D resin exchanging quickly and minimum requirement second investigated.
