Mass-Spectrometric MRM Analysis of FDA-Approved Proteins in Plasma of Healthy Volunteers

摘要: The proteomic composition of a biological sample serves as the most important feature object, and it allows discriminating normal pathological conditions. Targeted mass spectrometric analysis, particularly, multiple reaction monitoring (MRM) using synthetic stable isotope-labeled internal standards (SIS), is main alternative to ELISA method for analysis diagnostically significant proteins. Based on MRM results, prototype test system has been developed; employs targeted multiplex, quantitative FDA-approved proteins in whole plasma. Using this approach, was possible measure content 42 31 samples concentration range spanning five orders magnitude. interindividual variability 30 registered less than 40%. largest scatter observed haptoglobin (68%), immunoglobulin heavy constant delta IGHD (90%), angiotensin (72%), sex hormone-binding globulin SHBG (100%), lipoprotein-(a) (136%). obtained results correlate with published data (Hortin et al., Clinical Chemistry, 2008, vol. 54, 1608) R2 = 0.84. developed based can be considered an methods monoclonal antibodies.
