摘要: Specificity in immune responses is part controlled by the selective interaction of T cell receptors with their cognate ligands, peptide/MHC molecules. The discriminating nature this makes these molecules, soluble form, good candidates for selectively regulating responses. Attempts to exploit analogs proteins has been hampered intrinsic low avidity molecules ligands. To increase cognates biologically relevant levels, divalent complexes or (superdimers) were constructed. Using a recombinant DNA strategy, encoding either MHC class II/peptide TCR heterodimers was ligated coding murine Ig heavy and light chains. These constructs subsequently expressed baculovirus expression system. Enzyme-linked immunosorbant assays (ELISA) specific polymorphic determinants fraction molecule indicated that infected insect cells secreted approximately 1 νg/ml soluble, conformationally intact chimeric superdimers. SDS PAGE gel analysis purified protein showed expected molecular weight species. results flow cytometry demonstrated II chimeras bound specifically high bearing receptors. superdimers will be useful studying TCR/MHC interactions, lymphocyte tracking, identifying new antigens, have possible uses as regulators
