Cell-Type-Specific Cytokinin Distribution within the Arabidopsis Primary Root Apex

摘要: Cytokinins (CKs) play a crucial role in many physiological and developmental processes at the levels of individual plant components (cells, tissues, organs) by coordinating activities across these parts. High-resolution measurements intracellular CKs different tissues can therefore provide insights into their metabolism mode action. Here, we applied fluorescence-activated cell sorting green fluorescent protein (GFP)-marked types, combined with solid-phase microextraction an ultra-high-sensitivity mass spectrometry (MS) method for analysis CK biosynthesis homeostasis cellular resolution. This was validated series control experiments, establishing that protoplast isolation procedures did not greatly alter endogenous levels. The MS-based facilitated quantification all well known isoprenoid metabolites four transgenic Arabidopsis thaliana lines expressing GFP specific populations within primary root apex. Our results revealed presence gradient tip, concentration maximum lateral cap, columella, columella initials, quiescent center cells. distribution, when compared previously published auxin gradients, implies antagonistic interactions between two hormone groups are type specific.
