摘要: Background : Tetrahydrocurcumin (THC), an active metabolite of curcumin, has been reported to have similar biological effects but the mechanism antitumor activity THC is still unclear. Methods The present study was investigate and in human breast cancer MCF-7 cells using methods MTT assay, LDH flow cytometry analysis, western blot assay. Results found markedly cytotoxic effect antiproliferative against a dose-dependent manner with IC 50 for 24 h 107.8 μM. Flow analysis revealed that mediated cell-cycle arrest at G0/G1 phase, 32.8% entered early phase apoptosis 100 μM h. also dose-dependently led via mitochondrial pathway, as evidenced by activation caspase-3 caspase-9, elevation intracellular ROS, decrease Bcl-2 PARP expression, increase Bax expression. Meanwhile, cytochrome C released cytosol loss mitochondria membrane potential (Δψm) observed after treatment. Conclusion excellent source chemopreventive agents treatment be explored precursor compound.
