摘要: Genotoxic effects of air contaminants, such as gaseous or particulate compounds, have been difficult to investigate due inefficient methods for exposing cell cultures directly these substances. New cultivation and exposure techniques enable treatment epithelial cells with sample atmospheres subsequent in vitro assays, demonstrated by a new system called CULTEX (CULTEX: patent No. DE 19801763; PCT/EP99/00295), which uses transwell membrane technique direct complex mixtures, example sidestream cigarette smoke, at the air/liquid interface. The sensitivity susceptibility human bronchial this mixture already shown cytotoxic endpoints. In study, genotoxic smoke different concentrations were assessed using alkaline comet assay. HFBE 21 exposed 1 h clean air, nitrogen dioxide smoke. Exposure induced DNA strand breaks dose-dependent manner. combination gas phase assay provides realistic efficient model sensitive detection airborne inhalable compounds.
