摘要: The mammalian kidney filtration barrier is a complex multicellular, multicomponent structure that maintains homeostasis by regulating electrolytes, acid-base balance, and blood pressure (via maintenance of salt water balance). To perform these multiple functions, podocytes--an important component the apparatus--must process series intercellular signals. Integrating signals with diverse cellular responses enables coordinated response to various conditions. Although mature podocytes are terminally differentiated cannot proliferate, they able respond growth factors. It possible initial factors beneficial protective, might include induction hypertrophic cell growth. However, extended and/or uncontrolled factor signalling be maladaptive could result in apoptosis podocyte loss. Growth signal via activation receptor tyrosine kinases (RTKs) on their target cells around quarter 58 RTK family members encoded human genome have been identified podocytes. Pharmacological inhibitors many RTKs exist currently used experimental clinical cancer therapy. identification pathological RTK-mediated transduction pathways provide starting point for development novel therapies glomerular disorders.
