Effects of the Fusarium mycotoxins zearalenone and deoxynivalenol on the mitochondrial methylthiazol tetrazolium-cleavage activity of monolayer cells.

摘要: The application of a modified colorimetric assay for the evaluation mycotoxin-derived cytotoxicity is reported. Using four mammalian monolayer cell types (swine kidney, African green monkey Madin Darbin canine kidney and bovine embryonic lung cells), influence mycotoxins zearalenone deoxynivalenol on cellular methylthiazoltetrazolium (MTT)-cleavage activity was evaluated after different toxin exposure times. yellow tetrazolium salt converted by mitochondrial enzymes metabolically active cells into purple formazan products. These crystals were dissolved in dimethylsulphoxide to obtain homogeneous solution, optical density which suitable precise spectrophotometric measurement plate reader. Zearalenone proved be less cytotoxic than even enhanced cleavage tested. At concentrations 10 100 mug/ml, showed marked suppressive effect all types, depending length incubation time. results indicate that monolayers are useful targets MTT investigating action performing comparative studies cells.
