摘要: Tumstatin is a 28-kilodalton fragment of type IV collagen that displays both anti-angiogenic and proapoptotic activity. Here we show tumstatin functions as an endothelial cell-specific inhibitor protein synthesis. Through requisite interaction with alphaVbeta3 integrin, inhibits activation focal adhesion kinase (FAK), phosphatidylinositol 3-kinase (PI3-kinase), B (PKB/Akt), mammalian target rapamycin (mTOR), it prevents the dissociation eukaryotic initiation factor 4E (eIF4E) from 4E-binding 1. These results establish role for integrins in mediating inhibition cap-dependent synthesis suggest potential mechanism tumstatin's selective effects on cells.
