摘要: Structural aspects of lipoarabinomannans (LAM) from Mycobacterium tuberculosis and smegmatis were investigated by using mild acid hydrolysis in combination with Fourier-transform ion cyclotron resonance (FT-ICR), quadrupole trap mass spectrometry. Exact measurements less than 2.5 ppm error confirmed the presence a series arabinose oligomers (Aran; n=2–7) as major components observed following both M. LAM. However, spectrum resulting LAM extract also revealed highly-abundant distribution ions that exact identified mannose-linked species, AranManm+Na+ (n=1–6; m=1–3). The mannose caps linked to species mono-, di-, trimannose units, ratio was determined be 1.00:9.00:1.15, respectively, different previous reports. Analysis linkage lithiated trimer standards accomplished MS3 experiments information generated used identify linkages trimers spectra predominantly α(1 → 5), 5).
