Skewed X-chromosome inactivation and XIST locus methylation levels do not contribute to the lower prevalence of Parkinson's disease in females

摘要: Parkinson's disease (PD) is a degenerative disorder of the nervous system and cause majority sporadic cases unknown. Females are relatively protected from PD as compared with males linkage studies suggested susceptibility locus on X chromosome. To determine putative association skewed X-chromosome inactivation (XCI) PD, we examined XCI patterns using human androgen receptor gene-based assay (HUMARA) did not identify any or random clinical heterogeneity among female patients. In addition, sought to methylation-specific changes at X-inactive specific transcript (XIST) locus, which known be responsible for initiating inactivation. We observed trend towards hypomethylation in gene body region XIST females reach significance. Furthermore, extended our analysis DNA methylation across entire revealed no differences between healthy controls. Thus, propose that levels chromosome reveal could account decreased suitable use biomarker.
