摘要: Acetolactate synthase (ALS), the first enzyme in biosynthetic pathway of leucine, valine, and isoleucine, is biochemical target different herbicides. To investigate effects repression ALS activity through antisense gene expression we cloned an from potato (Solanum tuberosum L. cv Desiree), constructed a chimeric under control cauliflower mosaic virus 35S promoter, created transgenic plants Agrobacterium tumefaciens-mediated transfer. Two regenerants revealed severe growth retardation strong phenotypical resembling those caused by ALS-inhibiting Antisense decreased steady-state level mRNA these induced corresponding decrease up to 85%. This reduction was sufficient generate almost inviable without amino acid supplementation. In both herbicide-treated plants, could exclude accumulation 2-oxobutyrate and/or 2-aminobutyrate as reason for observed deleterious effects, but detected elevated levels free acids imbalances their relative proportions. Thus, inhibition generated vivo model herbicide action. Furthermore, RNA interest provides general method validation potential targets.
