摘要: A simple and reliable radioimmunoassay for plasma testosterone has been developed. Antiserum against was produced in rabbits immunized with testosterone-3-oxime-beef serum albumin. Plasma volumes of 0.1 ml from men 0.5 women were used analysis. After a preliminary solvent wash, the extracted extracts applied to Al2O3 micro columns. Following elution, dried purified extract incubated antiserum at room temperature 2 hours. (NH4)2SO4 separate free bound testosterone. 1,2-3H-testosterone correction losses determination % bound. The accuracy precision method satisfactory. sensitivity 10 pg per sample. mean blank ± (SD) Analysis duplicates by an alternative utilizing paper chromatography purification did not give results that differed significantly those obtained present method. range concentrations normal determined this similar reported other investigators using different methods.
