摘要: Publisher Summary This chapter outlines procedures for preparing mitoplasts and inner membrane vesicles assaying the import of precursors into these preparations. Import vesicles, such as intact mitochondria, requires an electrochemical potential across membrane. follows a protocol similar to that with modification buffer is supplemented ascorbate cytochrome c . The research on mitochondrial protein aims at identification proteins mediate precursor organelle. Several components are identified: some localized in outer membrane, others matrix. However, no has so far been shown be involved import. To identify proteins, biochemical systems allow study alone its manipulation methods needed. Reconstitution translocation activity proteoliposomes achieved E. coli export system microsomes from endoplasmic reticulum. Yeast provide starting point make available studying more detail.
