Direct Observation of H3–H4 Octasome by High‐Speed AFM

摘要: Despite evidence that histone H3 and H4 proteins may act as the precursor for orientating DNA sequence to form nucleosome structures, there is no direct of formed compact structure. Here, it demonstrated a H3-H4 octasome could be constructed without involvement H2A-H2B under in vitro reconstitution conditions. Atomic force microscopy was used obtain first observation structure, which exhibited similar core-protein size but with shorter core histone-binding region. The also displayed one-step histone-dissociation pattern heat treatment, distinct micrococcal nuclease peplomycin accessibility, suggests different wrapping nucleosomes.
