Detection of triclabendazole and three metabolites in bovine muscle samples with a gold nanoparticle-based lateral flow immunoassay

摘要: In this study, a highly sensitive monoclonal antibody (mAb) against triclabendazole (TCD) and its metabolites was produced by using novel hapten. The subclass of mAb IgG2a, IC50 values were 1.49–3.58 ng mL−1 TCD metabolites, affinity constant 5.46 × 109 mol L−1. We then went on to develop mAb-based lateral flow immunoassay (LFIA) for the rapid screening in foodstuff gold nanoparticles as label. LFIA-based test strip assembled routine procedures. All following parameters optimized including pH, minimum concentration needed synthesis AuNP–mAb conjugate, best coating antigen concentration, volume mAb–AuNP conjugate optimal diluent buffer constituents. Under conditions, cutoff LFIA 10 μg kg−1 TCD, TCD-SO TCD-SO2, 25 Keto-TCD, limits detection (LODs) 0.11 0.28 TCD-SO, 0.38 0.47 Keto-TCD. ranges quantitative 0.22–5.41 0.56–5.74 0.76–9.11 0.94–24.02 respectively. Finally, assay further applied detect real samples we are satisfied that AuNP-based is useful tool medicines samples.
