Characterization of neutral glycosphingolipids in human cataractous lens.

摘要: Neutral glycosphingolipids were purified from human senile cataractous lenses by a combination of solvent extraction, Folch's partition, acetylation, and column chromatography using DEAE-Sephadex Iatrobeads. Six major (A-F) monohexosylceramide to pentahexosylceramide identified sugar composition analysis, methylation secondary ion-mass spectrometry, glycosidase digestion, chromium trioxide oxidation. Their structures suggested that they closely related in their metabolism: chains sequence ceramide moieties similarly composed, namely C16:0 C24:1 constituted most the fatty acids, long-chain base components mostly C18-dihydrosphingosine with small portion C18-sphingosine. The implied two pathways branching lactosylceramide: one globotriaosylceramide other lactotriaosylceramide, which leads production Le(x) glycolipid via neolacto type 2 core chain.
