摘要: Abstract rRNA transcription in Xenopus laevis terminates near a 7-base-pair (bp) conserved sequence (T3 box) located 200 bp upstream of the site initiation for adjacent gene promoter. We present evidence here that 12-bp element containing T3 box is an essential part terminator. Using oocyte injection assay, we found (but not alone) severely reduced amount RNA detectable at sites downstream from itself and within was required to specify formation correct 3' ends. This requirement also seen pulse-label experiments by using homogenate nuclei, but data did allow us determine exact mechanism which acts. Removal region its normal location allowed significant readthrough transcripts accumulate, indicating additional sequences may be complete terminator function.
