摘要: Publisher Summary This chapter focuses on column liquid chromatography. The basic high-performance chromatography (HPLC) system consists of a pumping for solvent, an injection device to introduce analyte into the solvent flow, column, and detector. design pumps used in HPLC has changed little over years, although gradual improvements have made them more reliable easier use. operational requirements are ability deliver flow from 0.05–10 mL/min against back pressure up 400 bar. Large pump heads or dedicated needed high flows required preparative systems. majority systems include autosamplers. There number different designs, but most them, sample is collected vial with fixed needle/probe injected loop rotary valve, which then switched under computer control. An autosampler takes much operator variability out operation instrument, increases precision, enables virtually 24-hr equipment.
