摘要: Cytotoxic potential of melamine was evaluated with the use two in vitro models i.e. cell cultures rat hepatoma (line FaO) and skeletal muscle L6). The were exposed for 24, 48, 72 h to at eight concentrations, ranged from 0.01 10 mM. Four different assays applied which various biochemical endpoints assessed: mitochondrial activity - MTT reduction assay, proliferation Commassie Brilliant Blue (CBB) dye binding lysosomal neutral red uptake (NRU) membrane integrity LDH release assay. Effective concentrations (EC20, EC50 , EC80) calculated concentration-response curves, then they averaged over three independently conducted experiments. It found that assay most sensitive this compound. After 48 exposure values (mM, mean ± SD) FaO L6 cells 6.4 0.62, 8.2 1.51, respectively. inhibition measured by NRU damage plasma detected (but not cells; however, effects took place after longer (72 h) exposure. At time 5.2 mM 9.2 LDH, In spite low cytotoxicity melamine, more studies are needed hazard identification characterization
