摘要: We studied expression and functional characteristics of the insulin- insulin-like-growth-factor-I (IGF-I) receptors in human renal carcinoma. Ligand-binding properties tyrosine-kinase activity both receptors, as well 2 isoforms insulin receptor (HIR-A -B) were analyzed carcinoma normal adjacent kidney tissue 8 adult patients. Partially purified IGF-I from cell possessed identical affinities for their ligands. Renal carcinoma, however, contained 3- to 4-fold more specific insulin-binding sites 2-fold binding than tissue. In addition, we determined relative content insulin/IGF-I hybrids tissues. revealed similar amounts hybrids, i.e., 44 +/- 8.2% tracer 46 12.0% When equal protein studied, found significantly increased autophosphorylation elevated substrate phosphorylation To assess whether differences insulin-receptor caused by an altered pattern isoform expression, mRNA levels HIR-A -B. The were, expressed highly variable ratios tumor Our experiments show that expresses amount with each. increase cannot be explained isoforms.
