摘要: Plasma levels and serum protein binding of cis-diamminedichloro-platinum(II) (cis-DDP) or cis-diamminediaquoplatinum(II) (cis-aq) complexed to carboxymethyl-dextran (CM-dex) with a molecular weight 10,000 (T-10), 40,000 (T-40), 250,000 (T-250) were investigated in BALB/c mice. Levels active drug the circulation after i.v. i.p. administration free drugs, as well loss activity due following incubation mouse serum, monitored by an antitumor vitro assay using drug-sensitive tumor cell line. Following injection complexes, platinum(II) was maintained at higher for longer period, whereas disappeared rapidly. The rate disappearance from markedly influenced size carrier CM-dex, since retained amount considerably T-40 T-250 complexes than T-10 complex. An resulted rapid transient appearance low drugs blood, case transport slower their maintenance blood system higher. Serum much CM-dex-complexed (regardless CM-dex carrier) drugs.
