VIEW-MOD: A Versatile Illumination Engine With a Modular Optical Design for Fluorescence Microscopy

摘要: We developed VIEW-MOD (Versatile Illumination Engine with a Modular Optical Design): compact, multi-modality microscope, which accommodates multiple illumination schemes including variable angle total internal reflection, point scanning and vertical/horizontal light sheet. This system allows combining flexibly switching between different illuminations imaging modes by employing three electrically tunable lenses two fast-steering mirrors. versatile optics design provides control of 6 degrees freedom the source (3 translation, 2 tilt, beam shape) plus axial position plane. also standalone software an easy-to-use GUI to calibrate microscope. demonstrate applications this in biosensor imaging, optogenetics fast 3D volume imaging. is ready fit into complex circumstances requiring precise detection paths, has broad scope usability for myriad biological applications.
