Altered Oxidative Stress Responses in Nickel-resistant Mammalian Cells

摘要: BALB 3T3 cells exposed to NiCl2 acquired resistance concentrations as high 200 µm and retain for many generations in the absence of nickel. This was not due alterations uptake or metallothionein overexpression. The nickel-resistant B200 cell line found also exhibit cross-resistance hydrogen peroxide menadione. These had 1.8 times higher basal levels glutathione compared wild-type cells. Studies with synthesis inhibitor buthionine sulfoximine showed that while turnover more rapid cells, its depletion following treatment parental greater than reduced level binding NFkB AP-1 transcription factors their DNA consensus sequences reactive response resistant H2O2 sulfoximine, strengthens hypothesis nickel is closely allied oxidative stress responses.
