摘要: Abstract Objectives: The present study examines the effects of IL-1β and TGF-β1 in modulation ezrin, E-cadherin, CD44 β-catenin expression human trophoblast cells which may lead to their altered cytoskeleton dynamics during cell-to-cell cell-to-matrix interactions. Methods: Trophoblast (extravillous villous) isolated purified from early term placentae choriocarcinoma cell line JEG-3 used this were challenged with either or (10 ng/ml) for 12 h following RT-PCR was performed β-catenin. Immunolocalization these proteins carried out chorionic villi as well cultured stimulated by cytokines. Western Blot also regulation ezrin E-cadherin primary extravillous, villous Scanning electron microscopy (SEM) Matrigel Invasion Assay effect cytokines on cellular morphology invasion. Results: induced a down while upregulation message both cells. On contrary, exhibited just an opposite effect, i.e. up β-catenin, CD44. These observations further corroborated immunolocalization findings above first trimester tissue, former having predominance latter [Am. J. Reprod. Immunol. 48 (2002) 210]. Cellular observed through SEM revealed enhanced adhesion poor cell–cell interaction challenge strong intercellular weak presence TGF-β1. Crystal violet staining invasion higher index low challenge. Conclusion: mediated increased reduced along is associated invasiveness expression, invasiveness, morphology. facts therefore indicate possible role two motility alteration cell–matrix interaction.
