摘要: ABSTRACT Entamoeba histolytica is an intestinal protozoan parasite and the causative agent of amoebiasis. During invasive infection, highly motile amoebae destroy colonic epithelium, enter blood circulation, disseminate to other organs such as liver, causing liver abscess. Motility a key factor in E. pathogenesis, this process relies on dynamic actomyosin cytoskeleton. In systems, phosphatidylinositol 4,5-bisphosphate [PI(4,5)P2] known regulate wide variety cellular functions, including signal transduction, actin remodeling, cell motility. Little about role PI(4,5)P2 pathogenicity. study, we demonstrate that localized cholesterol-rich microdomains, lipid rafts, actin-rich fractions membrane. Microscopy revealed trailing edge polarized trophozoites, uroids, are enriched rafts their constituent lipid, PI(4,5)P2. Polarization enrichment uroids with were enhanced upon treatment cells cholesterol. Exposure cholesterol also increased intracellular calcium, which downstream effector PI(4,5)P2, concomitant increase Together, our data suggest histolytica, may from play triggering PI(4,5)P2-mediated signaling enhance motility pathogen.
