摘要: Two-dimensional column chromatography is a method which combines the advantages of (constant, adjustable flow velocity, excellent efficiency, on-line detection) and two-dimensional thin-layer (successive developments in two perpendicular directions, using different retention mechanisms). It shown that by merely keeping constant solvent velocity during development plate, considerable increase spot capacity can be achieved, since plate length particle size selected without any prejudicial influence on kept at value considered as optimum analyst. Calculations show generate peak capacities well excess 500, up to several thousands, specifications for equipment are not too stringent. A 10 × cm would 1 mm thick, packed with 10-μm particles operated reduced 10. Such packing could expected very homogeneous (A = 0.7) height 1.95. The 900, while pressure drop compound diffusion D 5 10-6, cm2/sec (linear u, 0.05 cm/sec) viscosity cP only atm (flow-rate 3 cm3/min). sample should about diameter or less. Equipment capable these performances under construction. Successful operation this separation scheme, however, relies ability find chromatographic systems operating according widely mechanisms.
