摘要: Publisher Summary Confocal scanning-laser microscopy combined with sophisticated image-processing techniques allow the cell biologist to explore three-dimensional infrastructure of cell, thus conferring a high degree precision in localization experiments. This chapter discusses some inherent problems preservation biological specimens so that reliable data can be acquired confocal fluorescence microscope. The methodology is result an effort study genesis epithelial polarity Madin-Darby canine kidney (MDCK) line. While specific methods have been developed these particular cells, lessons learned about their should widely applicable. A description principle introduces basic concepts microscopy. One then understand how images are recorded and information may interpreted.
