摘要: Abstract Background Propofol is a commonly used agent in total intravenous anesthesia (TIVA). However, the link between its pharmacokinetics and pharmacodynamics has not been fully characterized children yet. Our aim was to determine quantitative relationship venous plasma concentration bispectral index (BIS) effect heterogeneous group of pediatric patients undergoing various surgical procedures (ASA status I–III). Methods Nine male nine female were anesthetized with propofol–fentanyl TIVA. Sparse samples for propofol concentrations assay dense BIS measurements collected during after end infusion. Nonlinear mixed-effect modeling NONMEM data analysis. Results A three-compartment model linked classical E max through biophase compartment describe available data. All clearance volume terms allometrically scaled account body mass difference among under study. typical patient had their PK parameters observed within range literature values children. The pharmacodynamic highly variable. EC 50 2.80 mg/L distribution rate constant 3.33 min −1 found patient. Conclusions are correlated according concentration–response relationship. Children slightly lower sensitivity higher clearance, as compared adult literature. intra-patient variations require anesthesiologist's attention using alone evaluate depth
